You only need to use the Gain trim if your levels from your audio tracks or instruments aren't already where you want them. I prefer to get my levels as close to my reference level as possible from the source.

If you've been paying attention to your levels and they're already where they should be, I'd say don't touch the gain trims – grab them faders and start mixing! :-)
